Jyothika made her acting debut in the Hindi film Doli Saja Ke Rakhna, directed by Priyadarshan. She then starred in her first Tamil film Vaali (1999) and her first Telugu film Tagore, opposite Chiranjeevi. Her career gained momentum with the success of Vaali (1999) and Kushi (2000), and she received her first Filmfare Award for Best Female Debut – South for Vaali, and the Filmfare Best Tamil Actress Award for Kushi. She gained prominence for her performances in a string of successful films including Dhool, Thenali, Thirumalai, Dumm Dumm Dumm, Snegithiye, Kaaka Kaaka, Perazhagan, Chandramukhi, June R, Pachaikili Muthucharam, Mozhi, and 36 Vayadhinile. She received critical praise for playing a blind woman in Perazhagan, alter-egos Chandramukhi and Ganga in Chandramukhi, a mute and deaf girl in Mozhi, and a middle-aged woman in 36 Vayadhinile. At the peak of her career, she agreed to play the role of an antagonist in Pachaikili Muthucharam (2007). She left the industry at the peak of her career after marrying actor Suriya on 11 September 2006, with whom she was paired in seven films. She made a comeback in 36 Vayadhinile (2015), which received good reviews, especially for her performance, and she received the Filmfare Critics Award for Best Actress for the movie. She became the first actress to win both the Filmfare Award for Best Actress - Tamil and the Filmfare Critics Award for Best Actress. In recent years, she has appeared in films such as Naachiyaar (2018), Chekka Chivantha Vaanam (2018), Thambi (2019), Raatchasi (2019), Jackpot (2019), Ponmagal Vandhal (2020), and Srikanth (2024). She has also worked as a producer on films including Jai Bhim (2021), Sarfira (2024), Meiyazhagan (2024), and Retro (2025).
